Currently, the most mature route reported in the literature and applied industrially is the oxalyl chloride/DMF catalytic system, with anhydrous dichloromethane as the reaction solvent.

The specific procedure for this synthetic process is as follows: add anhydrous dichloromethane to a dried reaction flask, followed by the sequential addition of 3-cyclopentylpropionic acid and a catalytic amount of DMF. After thorough mixing, slowly add a 1.2-fold excess of oxalyl chloride under ice bath conditions, ensuring the reaction temperature does not exceed 10°C during the addition. Once the addition is complete, gradually raise the temperature to room temperature and continue stirring for 2–3 hours. After the reaction, remove the solvent and excess oxalyl chloride by distillation, then collect the target fraction via reduced-pressure distillation to obtain high-purity 3-cyclopentylpropionyl chloride.

This method offers notable advantages, with broad applicability at its core: it not only suits the acyl chlorination of 3-cyclopentylpropionic acid but also extends widely to preparing acyl chlorides from other aliphatic carboxylic acids—including both saturated and unsaturated types. It features mild reaction conditions, straightforward operation, and no need for specialized equipment. Furthermore, the purification process is simple and efficient, with distillation yielding high purity, thereby reducing the cost and complexity of industrial-scale production.

3-Cyclopentylpropionyl chloride serves as a key reagent in fragrance synthesis and the preparation of pharmaceutical intermediates. Its primary application lies in constructing carbonyl compounds containing cyclopentane structures through acylation reactions. Compared to traditional acylating agents, it offers high reactivity and excellent selectivity. When it reacts with carbon nucleophiles such as enolates or Meldrum’s acid, it rapidly accomplishes acylation under mild conditions and produces only hydrogen chloride as a by-product—one that is easily removable. Yields typically range between 80% and 90%.

In multi-step synthetic routes, this reagent serves as a critical acylating agent, effectively avoiding interference from unnecessary functional groups and enhancing the efficiency and atom economy of the synthesis. For example, in synthesizing fragrances with fresh fruity or floral notes, 3-cyclopentylpropionyl chloride can introduce cyclopentyl structures via acylation, enhancing the longevity of the fragrance.

In drug development, it can be used to synthesize intermediates for antibacterial or anti-inflammatory drugs featuring a cyclopentane core, providing a structural foundation for optimizing the activity of drug molecules.
